Glyoxalase I (Glo1) is responsible for cellular detoxification, specifically protecting cells from methylglyoxal (MG) toxicity. MG is a highly reactive carbonyl compound formed primarily as a byproduct of glycolysis. Due to MG’s extreme cytotoxicity, it spontaneously reacts with proteins, lipids, and DNA to form advanced glycation end-products (AGEs). Glo1 catalyzes the first step of the glyoxalase pathway, converting MG and reduced glutathione into S-D-lactoylglutathione, which is later converted to non-toxic D-lactate. Without adequate Glo1 activity, MG accumulation leads to profound damage of cellular infrastructure and induces oxidative stress and apoptosis. In diabetes, chronic hyperglycemia drives overproduction of MG resulting in diabetic complications such as nephropathy, retinopathy and neuropathy.
