Glycosaminoglycan (GAG) or mucopolysaccharide is a long, unbranched polysaccharides consisting of repeating disaccharide units. They are found in every mammalian tissue, components of the extracellular matrix and cell surface glycocalyx. With a high negative charge, GAGs are involved in structural integrity, cell signalling and hydration. The disaccharide consits of either N-acetylglucosamine or N-acetyl galactosamine and a uronic acid. With the exception of hyaluronic acid, GAGs are highly sulfated. They fall into 4 categories based on their disaccharide compostion: Hyaluronic acid, Heparin/heparin sulfate, chondroitin sulfate/dermatan sulfate and keratan sulfate. GAGs are typically attached to core proteins, forming proteoglycans. These have functions in diverse areas such as hydration and structure, cell signaling and growth, modulating cell adhesion, migration and proliferation. They play a key role in regulating tissue repair and inflammation. Alterations in GAG structure or amount are associated with atherosclerosis, cancer, neurodegenerative disorders and viral infections.
