Sulforhodamine B (SRB) cell cytotoxicity measurement is a widely accepted method of cell viability and drug cytotoxicity. It is based on the ability of basic amino acids in cellular proteins to bind SRB in TCA-fixed cells, providing a linear relationship between protein content (a proxy for cell number) and absorbance at 565 nm. The assay is independent of metabolic activity, eliminating drift and interference from various metabolic intermediates.
