3-D cell cultures are cell-culture systems where cells grow and interact in three dimensions, allowing cells to interact in a spatially complex environment, simulating real tissue architecture. This permits cell-cell and cell-matrix signaling which is not typically observed in traditional planar cell-culture systems. 3-D cell culture can occur using scaffolds such as hydrogels like Matrigel or with cells which self-assemble to form clusters. More advanced systems utilize bioprinting, miniature organoids or organ-on chip models trying to achieve the complexity of natural tissue environments. These 3-D cultures, however, make cell quantitation difficult due to variable responses to cellular assays. AkrivisBio’s 3DCell Culture Viability Assay allows for more accurate viability and cell quantitation. 3-D cell cultures previously grown by the user are gently disrupted and the cells are converted back to monolayers which are then measured for viability by detecting NADH leakage.
