DESCRIPTION

Aldo-keto reductases are a large superfamily of ~190 NAD(P)H-dependent oxidoreductases that catalyze the reduction of aldehydes and ketones to their corresponding alcohols in virtually all phyla of life. These enzymes play key roles in intermediary metabolism, detoxification of reactive carbonyl species, steroid and prostaglandin metabolism, and xenobiotic biotransformation. These are typically cytosolic monomers of about 37 kDa that adopt a conserved 8 -barrel (TIM-barrel) fold with characteristic surface loops that confer substrate specificity. A conserved tyrosine, histidine, and lysine form the catalytic tetrad together with an aspartate, enabling general acid-base catalysis during hydride transfer from NADPH or NADH to the carbonyl substrate. Overexpression or dysregulation can contributes to diabetic complications, cancer progression, and resistance to chemotherapeutic and antihormonal agents. Inhibitors and targeted degraders of isoforms such as AKR1B1 and AKR1C3 are under active investigation as therapeutic strategies in diabetic tissue injury and hormone-dependent malignancies, including prostate and hematologic cancers.
