Tumor necrosis factor-α-converting enzyme, also known as TACE and ADAM17, is a membrane-bound metalloprotease with several roles. It is the primary “sheddase” cleaving and releasing the extracellular domains of more than 80  membrane-bound proteins, including growth factors, cytokines and their receptors, allowing for a variety of cellular signaling mechanisms. Dysregulation of TACE is implicated in numerous diseases, including rheumatoid arthritis, atherosclerosis, IBD and cancer, and so is an interesting pharmacological target.
