Glycosylation of proteins is a common cellular mechanism to sort proteins for membrane attachment or secretion or to designate them for other specific purposes. O-linked mucin-type glycans are assembled, starting with the addition of N-acetylgalactosamine to a Serine or Threonine residue of a newly formed protein. This (Tn antigen) structure can be further modified by addition of either galactose, forming GalGalNAc (core1), sialic acid, forming sialyl-Tn or N-acetylglucosamine, forming GlcNAcGalNAc core3 structures, respectively. Core1 and core3 structures are then extended with the addition of further components, forming long, branched complex O-glycans.
