Cytidine deaminase (CDA) is an essential part of the pyrimidine salvage pathway, deaminating cytidine and deoxycytidine to uridine and deoxyuridine. This allows the recycling of nucleotides. CDA facilitates mitochondria biogenesis and oxphos and in some cancers where CDA activity is suppressed, mitochondrial respiration declines and glycolysis is increased, one of the classic biomarkers of cancer. Nucleoside analog drugs such as gemcitabine and cytaridine are rapidly inactivated by CDA giving rise to chemoresistance. Bloom syndrome is a condition where CDA deficiency or mutation results in replicative stress and genomic instability. CDA dysregulation also can result in immunodeficiencies and other metabolic disturbances.
