α-Glucosidase, primarily located in the brush-border of the small intestine, catalyzes the final step of starch and disaccharide digestion by hydrolyzing terminal, non-reducing α-1,4-glucoosidic linkages to release α-D glucose. This process is vital for the conversion of complex dietary carbohydrates into absorbable monosaccharides, thereby directly influencing blood glucose levels and overall energy homeostasis. A specific form, acid α-glucosidase is located in lysosomes. Deficiency in this enzyme is responsible for Pompe disease. Inhibition of this enzyme using acarbose or miglitol is an important method for managing type-2 diabetes and are being explored for weight management and cardiovascular risk factors.
