Phospholipases A2 are a family of enzymes that catalyze the hydrolysis of the sn-2 fatty acid of a glycerophospholipid releasing a lysophospholipid and the free fatty acid. The PLA2 superfamily can be divided into three major types based upon size and cation dependence. Two subtypes, the low MW secretory enzymes (sPLA2) and the high MW cytosolic enzymes (cPLA2), require Ca2+ for activity. sPLA2 enzymes play an important role in the digestion of dietary phospholipids, defend against bacterial infections and release free eicosanoids. cPLA2 enzymes are involved in in inflammation, releasing polyunsaturated fatty acids from membrane lipids allowing production of a host of oxidized eicosanoids. The third type, high MW CA2+-independent enzymes (iPLA2), are intracellular enzymes that appear to be involved in phospholipid remodeling, cell proliferation and fat catabolism.
