6-Phosphogluconate (6PG) is a metabolic intermediate in the oxidative branch of the pentose phosphate pathway, the substrate for 6-phosphogluconate dehydrogenase. It sits at the intersection of carbon homeostasis, reducing equivalent production (NADPH), and anabolic biosynthesis. Its accumulation serves as a regulatory signal affecting glycolysis and cell signaling. As an inhibitor of gluose-6-phosphate isomerase, it forces the flow of glucose into the pentose phosphate pathway to address high anabolic demands or oxidative stress. It links carbohydrate metabolism to protein secretion as its suppression causes endoplasmic reticulum dilation due to increased extracellular matrix protein congestion.
