6-Phosphogluconate dehydrogenase a step in the hexose monophosphate shunt and pentose phosphate pathways, catalyzes the oxidative decarboxylation of 6-phosphogluconate to ribulose-5-phosphate with reduction of NADP to NADPH. Ribulose-5-phosphate is a precursor of nucleotides and nucleic acids and is reversibly converted to glyceraldehyde-3-phosphate and fructose-6-phosphate to enter glycolysis. It is an important source of NADPH, critical for biosynthetic pathways and prevention of oxidative damage. In humans, its deficiency results in depletion of NADPH in red blood cells. 6-PGD deficiency increases the risk of hemolytic anemia during oxidative stress. Inhibition of the enzyme shows anti-cancer activity, decreasing RNA biosynthesis and increasing ROS in tumor cells.
