Oxalate Decarboxylase, a member of the cupin superfamily, catalyzes the removal of CO2 from oxalate forming formate. In humans, oxalate is a metabolic end product, cannot be further metabolized and is either metabolized by gut microorganisms or excreted, primarily through the kidneys. It is mildly toxic and causes hyperoxaluria and when combined with calcium, forms kidney stones which can lead to kidney failure. In microorganisms, oxalate decarboxylase helps maintain cellular homeostasis. In fungi, it is involved in morphogenesis, stress resistance and in some cases, virulence, since oxalate degradation modulates the local pH influencing host-pathogen interactions. It is being considered as a potential therapeutic treatment for hyperoxalemia and hyperoxaluria.
