Neuraminidase hydrolyzes terminal sialic acid residues from glycoproteins, glycolipids and oligosaccharides, which contribute to a range of physiological and pathological processes. In mammals, 4 main types of neuraminidases (NEU1-4) display different tissue distributions, subcellular locations and substrate specificities. Neuraminidases regulate metabolism by affecting glycoprotein and glycolipid turnover in the lysosome. NEU1 modulates cell receptors by desialylating them which can change receptor activity and downstream signaling pathways. Neuraminidase dysfunction appears to be involved in a variety of disease state from some lysosomal storage diseases, type 2 diabetes, neurodegenerative disorders, some cancers, cardiovascular disease and respiratory pathologies.
