β-Glucuronidase is a lysosomal enzyme which hydrolyses β-D-glucuronic acid residues from a range of drugs, toxins and other xenobiotics. Phase II detoxification includes the conjugation of glucuronic acid to a variety of molecules, making them more water soluble and easily excreted. β-glucuronidase can reverse this with detrimental results. Bacterial β-glucuronidase in the gut affects enterohepatic circulation of many compounds. It also affects the balance of active vs. inactive hormones such as estrogen with implications for hormone-related conditions such as some cancers. Due to its effect on detoxification, hormone regulation and drug metabolism it is being explored as a target for drug development.
