Aldolase catalyzes the reversible transformation of fructose-1,6-biphosphate to glyceraldehyde-3-phosphate and dihydroxyacetone phosphate, a key step in glycolysis and gluconeogenesis. Aldolase acts as a metabolic switch during glycolysis and gluconeogenesis regulating glucose homeostasis. Aldolase has 3 forms, A, B and C localized in different tissues, A primarily in muscle, B in liver and C in brain, each with distinct physiological roles. Dysregulation of aldolase is seen in cardiovascular and neurodegenerative disease as well as in inherited metabolic disorders (fructose intolerance) and cancer where upregulation supports heightened glycolytic activity (Warburg effect).
