Cytochrome P450 3A4 is the predominant form of P450, with approximately 30-50% of the total P450 activity in the liver. It is responsible for the phase 1 biotransformation of an extremely wide range of xenobiotics as well as many endogenous compounds. Inhibition of this isoform is responsible for a number of adverse drug reactions and drug/drug interactions. AkrivisBio’s CPY3A4 Assay is a simple, sensitive assay for enzyme activity using a sensitive substrate which is converted to 7-Hydroxy-4-trifluoromethylcoumarin with a dramatic increase in fluorescence at 385/500 nm. Specificity is provided by the addition of ketoconazole to the kit which is an index inhibitor of CYP3A4.
