lutathione is the principal intracellular low-molecular-weight thiol which maintains the redox state of the cell. It balances the constant oxidative metabolism of living systems, neutralizing the appearance of reactive oxygen and nitrogen species. It is the primary cellular defense against oxidative stress in mammalian cells. AkrivisBio’s Glutathione (DTNB) Assay provides a simple convenient way to quantify either total (oxidized + reduced) glutathione or reduced glutathione only. The assay takes place in two steps. The first step, utilizing DTNB, reacts with reduced glutathione converting it to GSSG, reducing DTNB to monomeric thiol form with intense color. Using this step alone allows for quantitation of reduced glutathione (GSH) only in an endpoint assay. The second step utilizes glutathione reductase and NADPH to convert any oxidized glutathione (GSSG) to the reduced form which is then available to react with DTNB. This results in a highly sensitive recycling assay for glutathione as shown in the figure below. Sensitivity is below 1 ng of glutathione using the recycling reaction and about 100 ng of glutathione using the endpoint reaction.
